Some people say one should allow around three months before you begin to check the watch's accuracy. While I dont believe watches run in like cars, there is some substance to the observation that a run in period allows the lubricant to properly settly, particularly if its been in store or immobile.
The rotor is a bi-directional rotor. Generally, all you need to do is wind it up a few turns and let the rotor do its job of winding the watch.
Watch winders are fine, but it wont really hurt your watch leaving it unwound until weekends. Its a minor issue to change the date settings, but if you wish to keep it in a winder there is no problem with that.
